EPC room in roof

Hi hoping someone can help me out in explaining what exactly is rooming roof insulation. I have a HMO and it has a room in the loft.

Does room in the roof insulation only apply to the attic room or could I increase my EPC rating by putting insulation between the ground floor and the first floor floorboards? Example does room in the roof insulation apply to having insulation above the lounge?

That will not change your EPC.

In broad terms the EPC considers the energy input to, and the heat loss from, the building as a whole.

Heat flow from one room to another, whether on the same floor, or between floors, is not a factor in the calculation.

It means insulation to reduce heat loss from the attic room to the outside world via its the room's walls and ceiling.
